ABSTRACT

Objective:To establish a HPLC method for determination of chlorogenic acid,rutin,polydatin,kaempferol-3-O-rutinoside,astragalin,resveratrol,quercetin,kaempferol in Tetrastigmatis Hemsleyani Radix,in order to study the content changes of eight components of different months. Method:Zorbax SB C18column (4.6 mm×250 mm,5 μm)was adopted with mobile phase consisting of acetonitrile(A)and 0.1%phosphoric acid(B)in gradient elution(0-30 min,10%-30%A;30-40 min,30%-95%A;40-45 min,95%A;45-60 min,95%-10%A). The flow rate was 0.8 mL·min-1,the column was kept at 25℃,and the detection wavelength was 320 nm. Result:Chlorogenic acid,rutin,polydatin,kaempferol-3-O-rutinoside,astragalin,resveratrol,quercetin,kaempferol showed a good linearity within the range of 13.7-549 mg·L-1(r=0.999 0),12.6-253 mg·L-1(r=0.999 1),15.8-316 mg·L-1(r=0.999 0),14.7-147 mg·L-1(r=0.999 2),8.8-88 mg·L-1(r=0.999 1),7.9-79 mg·L-1(r=0.999 5),8.6-172 mg·L-1(r=0.999 1),8.9-89 mg·L-1(r=0.999 4). There were great differences in contents of the eight flavonoid active components in different growth phases. In July and August, the relative contents of chlorogenic acid, rutin, kaempferol 3-O-rutinoside and polydatin were the highest. The highest relative content of quercetin was observed in June. The relative contents of resveratrol and kaempferol in April and May was higher than those in other mouths. The relative content of astragalin in November was the highest. Conclusion:It could provide abundant information for the production and quality control of Tetrastigmatis Hemsleyani Radix.

ABSTRACT

Objective To analyze the characteristic changes of trace elements from Hedysari Radix in different months; To provide references for the choice of the best harvest periods. Methods Trace elements of Hedysari Radix from different months (June, July, August, September, October and November) were determined by atomic absorption spectroscopy and analyzed by single factors analysis of variance, correlation analysis, cluster analysis and factors analysis. Results There was obvious significance between the contents of Cu and Co (P<0.05), and there was obvious significance among the contents of Fe, Ca, Mn, Zn, Mg, Cr, Na, Li, Ni and K (P<0.01). The results of correlation analysis showed that months had positive correlation with Cu, had a negative correlation with Fe, Na, Mn, Cr and Li contents, and had significantly negative correlation with Ca, Mg contents. Mn, Cr, Ni, K and Na were the characteristics of trace elements from Hedysari Radix in different months. The different months samples could be divided into two categories of June, July, August, September samples (Ⅰ) and October, November samples (Ⅱ). Conclusion There is close relationship between the contents of trace elements from Hedysari Radix and months. The information and metabolic patterns of trace elements from Hedysari Radix in June, July, August, September (immature) samples are close or similar;the information of trace elements in October, November (nearly mature or maturing) samples are close or similar. Study on harvest times of Hedysari Radix by trace element as an indicator is with certain rationality.

ABSTRACT

AIM: To investigate timed changes of bone histomorphometry parameters in normal rats of different months and to provide a control prove for research of drugs of antiosteoporosis. METHODS: Thirty two female rats at aged 4.5 months were divided by their weight ( 256.3 ? 25.3 g). Tetracycline derivates and calcein were subcutaneous injected to each rat on two separate occasions where labeled the sites of bone formation. All rats were sacrificed at 0, 30, 75 and 140 days. The undecalcified longitudinal proximal tibial metaphyseal sections were cut and stained for quantitative bone histomorphometriy. RESULTS: The cancellous bone mass increased slowly and then decreased, but there were no significant differences between 4.5 and 10 months. Bone mass kept relatively stable. Both bone formation and bone resorption increased first and then decreased, but there was a significant difference between 4.5 and 10 months, and bone turnover kept stable. CONCLUSION: The body weight and the bone mass parameters keep relatively stable in SD rats during the 4.5 to 10 months, and these rats can be selected as the models for the study of the drugs of antiosteoporosis.
